Gram Stain

The Gram Stain is used for differential staining of bacteria from cultures or patient samples.

The staining behaviour is an important taxonomical attribute, which is also correlated with other bacterial characteristics.

The specimen is first stained with Gentian Violet and then fixed with Iodine Solution. The absorption of the stain and of the complex formed between Iodine and Gentian Violet in gram-positive and gram-negative bacteria are similar, however the solubility of the Stain-Iodine complex is different. Gram-positive bacteria retain the Stain-Iodine complex when treated with Decolorizer Solution and remain blue. Gram-negative bacteria are decolorized and recoloured by staining with Safranin.
